FDA Imposes Full Clinical Hold on Aardvark's PWS Drug Candidate, Triggering Sharp Stock Decline

Regulatory Blow to Core Pipeline The U.S. Food and Drug Administration has issued a full clinical hold order for Aardvark Therapeutics' core investigational drug ARD-101, freezing all ongoing clinical trials for Prader-Willi syndrome. Impacted by this news, the company's stock plummeted over 30% in Friday's pre-market trading, closing down approximately 24%. Year-to-date losses have now exceeded 60%.

Safety Signals Prompt Concern This regulatory action stems from cardiac safety concerns identified in a healthy volunteer study. Analysis revealed reversible cardiac electrical activity abnormalities in subjects receiving the 1600 mg dose, which may indicate reduced cardiac pumping efficiency. Aardvark had proactively paused the related study as early as February this year, but the FDA's latest decision has cast a more pessimistic outlook on the program's future.

Strategic Contraction and Data Unblinding The hold affects not only the Phase III HERO study for Prader-Willi syndrome and its open-label extension study but also trials for another obesity candidate, ARD-201, which shares the same mechanism of action. In search of a path forward, Aardvark plans to unblind and analyze the accumulated Phase III clinical data to assess the overall safety and efficacy profile of ARD-101.

Financially Sound but a Challenging Road Ahead The company reiterated its solid financial position, holding approximately $91.2 million in cash and investments as of the end of the quarter, sufficient to fund operations into mid-2027. However, several investment banks, including Morgan Stanley, have swiftly downgraded the stock, noting this event introduces significant uncertainty regarding the program's prospects.
