CSPC PHARMA’s ADC Candidate SYS6043 Earns NMPA Breakthrough Therapy Status for Platinum-Resistant Ovarian Cancer

CSPC Pharmaceutical Group Limited (CSPC PHARMA) announced that its self-developed B7-H3–targeting antibody-drug conjugate, SYS6043, has been granted Breakthrough Therapy Designation by China’s National Medical Products Administration. The designation covers monotherapy treatment of platinum-resistant ovarian cancer, primary peritoneal carcinoma and fallopian tube cancer.

Ovarian cancer remains a high-burden malignancy with significant unmet need: approximately 0.32 million new cases were reported globally in 2022, and over 70% of patients present with advanced disease, resulting in a five-year survival rate of about 40%. Among those receiving platinum-based chemotherapy, 70%–80% experience disease progression. Current non-platinum chemotherapy regimens deliver an objective response rate of only 10%–15%, a duration of response of 3–4 months and median overall survival of roughly 12 months.

In clinical studies, SYS6043 demonstrated a progression-free survival roughly twice that achieved by existing non-platinum monochemotherapy and mirvetuximab soravtansine, while maintaining a favourable safety profile. CSPC PHARMA has commenced a Phase III confirmatory trial in the designated indication and is simultaneously evaluating SYS6043 in first- and second-line small-cell lung cancer, breast cancer and additional solid tumours.

The Breakthrough Therapy Designation is expected to expedite SYS6043’s development, review and potential market entry, broadening CSPC PHARMA’s oncology portfolio and reinforcing the group’s strategic position in antibody-drug conjugates.
