RiboLife-B (06938) has unveiled new Phase IIa data for its core candidate, Vortosiran (RBD4059), at the CPIC conference, reinforcing the drug's profile as a potent, durable, and safe therapy. The data showed an average maximum reduction of FXI activity of 92%, with suppression lasting for several months, alongside an excellent safety profile.

Groundbreaking Innovation Gains Further Validation

As the world's first and fastest-clinically-advancing FXI siRNA drug for thrombotic diseases, vortosiran is developed using RiboLife's proprietary RiboGalSTAR™ liver-targeting platform. It works by specifically inhibiting FXI expression and blocking the activation of the intrinsic coagulation pathway, achieving precise anticoagulation and antithrombotic effects. The new Phase IIa data for the coronary artery disease (CAD) indication shows that in patients who completed the high-dose regimen with a maintenance dose of 400mg, the average maximum reduction in FXI activity reached 92%, with effects persisting for months post-dosing. This supports a potential dosing regimen of once every 3 to 6 months across different indications. Regarding safety, vortosiran demonstrated a favorable profile in the Phase IIa trial, with no treatment-related serious adverse events, major bleeding events, or clinically relevant non-major bleeding events reported. Compared to other investigational drugs targeting the same pathway, vortosiran shows a significant advantage, with its FXI inhibition markedly higher than the estimated levels for small molecule drugs in Phase III development, which typically require once- or twice-daily dosing. Notably, these results provide the world's first clinical proof-of-concept for an FXI siRNA in coronary artery disease patients, supporting vortosiran's potential as a differentiated, long-acting antithrombotic therapy. By targeting FXI, vortosiran aims to reduce thrombotic risk while preserving the body's hemostatic function, potentially addressing a major unmet medical need in cardiovascular disease and further validating its pioneering, game-changing innovation value.

Aiming at a Massive Global Market

While antiplatelet and anticoagulant therapies are standard clinical practices, the risk of thrombotic events remains inadequately controlled in many patients with stable coronary artery disease (CAD) even after optimized standard treatment. A major limitation of existing therapies is their high bleeding risk, which restricts treatment intensity and long-term regimens for many patients. The Phase IIa results for vortosiran validate its potential to address this antithrombotic treatment dilemma in CAD, demonstrating the ability to provide effective, long-lasting prevention of thromboembolism while controlling bleeding risk, thereby balancing efficacy and safety. From a market perspective, thromboembolic diseases are implicated in approximately one-quarter of global deaths annually. The global antithrombotic drug market is projected to reach $70 billion by 2026 and exceed $100 billion by 2031. It is estimated that about 55% to 60% of this market could be targeted by FXI-targeting siRNA therapies, indicating a vast unmet demand. Targeting this critical global need for precise anticoagulation, RiboLife has a long-term strategic plan. According to the latest R&D progress, the company submitted two Phase IIb clinical trial applications to the European Medicines Agency (EMA) in the second quarter of this year for stroke prevention in atrial fibrillation (SPAF) and venous thromboembolism (VTE). These milestones represent a key breakthrough in the clinical development of small nucleic acid drugs in the global anticoagulation therapy field. In its clinical development strategy for the anticoagulation market, RiboLife is not only pursuing a "saturation attack" across multiple indications with vortosiran but has also established a comprehensive "combination" approach. For instance, in May of this year, RiboLife submitted a Clinical Trial Application (CTA) to the EMA for a Phase II trial of RBD1119, another small nucleic acid drug for treating CAD. This fully demonstrates the company's differentiated development strategy and strong determination to conquer the cardiovascular field, with its leading global position in anticoagulant/antithrombotic small nucleic acid therapeutics becoming increasingly clear. The upcoming readouts of key data and potential approvals for several of the company's other major anticoagulant drugs will serve as decisive validation points for RiboLife's multi-product portfolio to synergistically drive growth, leverage the multi-billion dollar global anticoagulation market, and solidify its pioneering leadership position.
