Alphamab Oncology (09966.HK) Initiates Phase I Trial, First Patient Dosed with PD-L1/VEGFR2 Bispecific ADC JSKN027

Alphamab Oncology (Stock Code: 09966), a PRC-based biopharmaceutical group, announced the successful dosing of the first patient in a Phase I clinical study of JSKN027, the industry’s first PD-L1/VEGFR2 bispecific antibody-drug conjugate (ADC) to reach human trials.

JSKN027-101 is an open-label, multi-centre Phase I study encompassing dose-escalation and dose-expansion stages. Key objectives include determining the maximum tolerated dose and recommended Phase II dose while evaluating safety, tolerability, pharmacokinetics/pharmacodynamics and preliminary anti-tumour activity in patients with advanced malignant solid tumours.

JSKN027 represents Alphamab’s fifth ADC to enter clinical development. Built on the company’s glycan-specific conjugation technology, the molecule links a topoisomerase I inhibitor payload to a bispecific antibody that simultaneously targets PD-L1 and VEGFR2. The mechanism combines targeted chemotherapy, anti-angiogenesis and immune-checkpoint blockade, aiming to enhance anti-tumour efficacy and overcome resistance across multiple solid tumour types.

Alphamab operates an integrated R&D platform spanning ADCs, bispecific antibodies and multifunctional protein engineering. Its pipeline comprises one NMPA-approved product alongside several Phase III or pivotal-stage assets. The company cautions that clinical development outcomes are uncertain and there is no guarantee of eventual market approval for JSKN027.
