Sanofi's Development Pipeline Stumbles Again as Rare Disease Drug Trial Halts, Adding Pressure on New CEO

The French pharmaceutical giant Sanofi SA (SNY.US) announced on Wednesday that it is ending a late-stage clinical trial early for an experimental drug targeting a rare autoimmune disease, as the treatment failed to demonstrate effectiveness. This latest setback increases the operational pressure on recently appointed Chief Executive Officer Belén Garijo.

Key Trial Halts, Riliprubart's Future in Question

Sanofi stated it will stop the study of riliprubart for treating chronic inflammatory demyelinating polyneuropathy (CIDP), a rare autoimmune disorder that causes progressive limb weakness and sensory disturbances, severely impacting patients' quality of life. The company said it will evaluate whether to continue developing the drug for other potential uses. Riliprubart was previously seen as a core asset in Sanofi's rare disease pipeline and a key candidate analysts expected to report pivotal data this year. This failure disrupts the company's planned growth transition to new products.

Successive R&D Setbacks Compound New CEO's Challenges

Garijo took the helm this past February under difficult circumstances, following the ouster of former CEO Paul Hudson for failing to bring new drugs to market quickly enough. Garijo's primary task is to build a stronger foundation of new products before the company's blockbuster drug Dupixent faces a patent cliff and diminishing revenue growth. However, this trial failure is not an isolated incident. Sanofi has recently encountered challenges in several key development programs: amlitelimab, an experimental drug for atopic dermatitis (eczema), showed mixed clinical data and failed to demonstrate a consistently reliable efficacy advantage; and tolebrutinib, an oral BTK inhibitor for multiple sclerosis. Despite receiving a recommendation for approval in Europe, it has faced delays in gaining U.S. regulatory clearance, raising market doubts about its global prospects.

Dupixent Patent Cliff Countdown

Sanofi's current primary growth engine, Dupixent (dupilumab), is set to face the dual pressures of biosimilar competition and patent expiration in the coming years. Investors are closely watching whether the company can develop sufficiently substantial follow-up products within this critical window. With the Phase III failure of riliprubart, market confidence in Sanofi's R&D capabilities is likely to be further tested. Analysts note that new CEO Garijo must quickly revitalize the development pipeline and improve clinical success rates; otherwise, the company may struggle to smoothly navigate the anticipated earnings downturn caused by the Dupixent patent cliff.
