专利摘要显示，本发明公开了一种利用click reaction合成环状RNA的方法。本发明提供合成环状RNA的方法，包括如下步骤：1)以待环化线性DNA为模板合成RNA，且在所述RNA的两端添加炔基修饰和叠氮修饰，得到两端修饰炔基和叠氮的RNA；2)将所述两端修饰炔基和叠氮的RNA在Cu离子的催化下进行点击反应，收集点击反应产物，即得到环状RNA。本发明通过点击化学，在反应结束后，加入固相叠氮或炔基标记物，能大大简化纯化过程，提高纯化效率；本发明通过点击化学，只需通过序列互补，将首尾拉近，即可进行反应，条件可控且反应效率高。因此，可大大提高环化效率。
