智通财经APP讯，加科思-B(01167)发布公告，公司自主研发的泛KRAS抑制剂JAB-23E73在美国完成I/IIa期临床试验首例患者给药。目前该药物在中国的剂量递增研究也正按计划顺利推进。
KRAS是最常导致致癌突变的基因之一，约23%-25%的癌症患者带有KRAS突变，其在胰腺癌(约90%)、结直肠癌(约40%)和非小细胞肺癌(约30%)中突变频率尤为显著。全球每年新增约270万患有KRAS相关突变的肿瘤患者，这突显了该患者群体巨大的未满足医疗需求以及泛KRAS抑制剂可能带来的潜在益处。
JAB-23E73对KRAS具有高度选择性，可同时抑制活性及非活性状态的KRAS，且对 HRAS、NRAS无明显抑制。作为一款口服KRAS抑制剂，JAB-23E73在临床前研究中展现出良好的药代动力学特征和显著的抗肿瘤活性。
