阿斯利康今日公布，其潜在同类首创CLDN18.2靶向抗体偶联药物Sone-Ve在III期CLARITY-Gastric01试验中达到主要终点，与研究者选择的治疗方案相比，在二线及后线CLDN18.2阳性晚期胃癌患者中实现了具有统计学显著性和高度临床意义的总生存期改善。

　　该试验纳入局部晚期或转移性胃癌、胃食管结合部癌或食管腺癌患者，要求肿瘤细胞CLDN18.2表达≥25%。试验达到双重主要终点之一——三线及后线治疗中的总生存期，同时关键次要终点——二线及后线总体人群的总生存期也显示出显著改善。不过，另一主要终点无进展生存期虽呈改善趋势，但未达到统计学显著性。

　　在安全性方面，Sone-Ve耐受性良好，与已知安全性特征一致，未发现新的安全信号。

　　CLDN18.2已成为胃癌的重要治疗靶点，约60%的胃食管癌患者肿瘤细胞表达该蛋白。晚期胃癌患者预后极差，一线治疗进展后中位生存期仅5-9个月。阿斯利康肿瘤研发执行副总裁Susan Galbraith表示，Sone-Ve有望通过新型靶向ADC替代传统化疗，重塑胃癌治疗格局。详细数据将在即将召开的医学会议上公布并递交监管机构。
